Analyte InformationAmino acid sequence 21-167 of human ZG16p (with the single substitution Y104F) was expressed in Escherichia coli using pCold- glutathione S-transferase (GST) vector. The ZG16p is a soluble mammalian lectin with a Jacalin-related b-prism-fold. This mutant protein was predicted to show diminished binding to a-mannose-related sequences compared to the wild-type ZG16p.

ProtocolAfter blocking arrayed slides with 3% (w/v) bovine serum albumin (Sigma A8577) in PBS, GST-ZG16p (Y104F) was overlaid onto the arrays at 200 ug/ml (undiluted) followed by rabbit anti-GST antibody Z-5 (Santa Cruz), 1:200, and then by biotinylated anti-rabbit IgG (Sigma), 1:200. Binding was detected using Alexa Fluor-647-labeled streptavidin from Molecular Probes (1 ug/ml).
