Analyte Information	The VP8* domain (amino acids 46 to 231) of human rotavirus P[19] (GenBank accession number DQ887060) was expressed in E. coli BL21 and purified by glutathione S-transferase (GST) tag affinity purification. It has been known that the VP8* domain of the VP4 spike protein plays a key role in rotavirus attachment to cell surface carbohydrates, an important first step for infection.
Concentration	50 ug/ml
Protocol	After blocking arrayed slides with 0.02% (v/v) Blocker Casein (Pierce), 1% (w/v) bovine serum albumin (Sigma A8577) in HEPES-buffered saline (5 mM HEPES, pH 7.4, 150mM NaCl, 5 mM CaCl2), the P[19] VP8* was overlaid at 50 μg/ml, followed by rabbit anti-GST antibody Z-5 (Santa Cruz) at 1:200 dilution and biotinylated anti-rabbit IgG (Sigma) at 1:200 dilution. Bnding was detected using biotinylated anti-rabbit IgG (Sigma) at 1:200 dilution followed by Alexa Fluor-647-labeled streptavidin from Molecular Probes (1 ug/ml). As diluent the blocker solution was used.
