Analyte InformationRecombinant DC-SIGNR (complete extracellular domain lacking the transmembrane domain) was recombinantly expressed in Escherichia coli (Mitchell et al. 2001, J Biol Chem 276: 28939-28945) and analysed in the microarrays in a biotinylated form.

ProtocolAfter blocking arrayed slides with 0.02% Blocker Casein (Pierce) containing 1% (w/v) bovine serum albumin (Sigma A8577) and 5 mM CaCl2 in Hepes buffered saline (5 mM Hepes, pH 7.4, 150 mM NaCl), DC-SIGNR-biotin was analysed at 50 μg/ml. Binding was detected using Alexa Fluor-647-labeled streptavidin from Molecular Probes (1 ug/ml). As diluent the blocker solution was used.
