Analyte InformationExtracellular domain of DC-SIGN (amino acid residues 64-404) fused at the C terminus to a human IgG1-Fc fragment and recombinantly expressed in CHO cells (DC-SIGN-Fc). DC-SIGN is a signalling C-type-lectin receptor of the immune system that plays a role in cell-cell and host-pathogen interactions.

ProtocolAfter blocking arrayed slides with 0.02% (v/v) Blocker Casein (Pierce), 1% (w/v) bovine serum albumin (Sigma A8577) in Hepes buffered saline (5 mM Hepes, pH 7.4, 150 mM NaCl, 5 mM CaCl2), Fc-DC-SIGN was assayed at 2 ug/ml, followed by biotinylated anti-human IgG (Vector) at 10 ug/ml. Binding was detected using Alexa Fluor-647-labeled streptavidin from Molecular Probes (1 ug/ml). As diluent the blocker solution was used.
