Analyte Information	Engineered chimeric human-murine antibody derived from the anti-beta-glucan mAb 2G8. The VH and VL regions of murine mAb 2G8 were assembled in a Single chain (Sc) variable fragment (scFv) format and further fused to the Fc-encoding sequence of the human IgG1 gamma chain (including hinge, CH2 and CH3 domains). The genes were recombinanlty expressed using an Agrobacterium tumefaciens-mediated transient expression system in Nicotiana benthamiana plants. The recombinant chimeric 2G8 scFvFc antibody was shown to retain the antifungal activities of the original murine mAb 2G8 against Candida albicans.

Protocol	After blocking arrayed slides with 0.02% (v/v) Blocker Casein (Pierce), 1% (w/v) bovine serum albumin (Sigma A8577) in Hepes buffered saline (5 mM Hepes, pH 7.4, 150 mM NaCl, 5 mM CaCl2), chimeric 2G8 scFvFc was assayed at 0.5 ug/ml, followed by biotinylated anti-human IgG (Vector) 1:200. Binding was detected using Alexa Fluor-647-labeled streptavidin from Molecular Probes (1 ug/ml). As diluent the blocker solution was used.
