2012 Nov 20
Analyte NamePGT 121
Analyte FamilyAntibodies
Analyte InformationOne of the human HIV-1 broad and potent neutralizing IgG antibodies isolated from HIV-1-infected individuals by single-cell sorting and neutralization screen and appears to bind to an Asn332 glycan-dependent epitope on gp120.
Concentration50 ug/mL
ProtocolAfter blocking arrayed slides with 0.04% (v/v) Blocker Casein (Pierce), 1% (w/v) bovine serum albumin (Sigma A8577) in Hepes buffered saline (5 mM Hepes, pH 7.4, 150 mM NaCl, 5 mM CaCl2), PGT121 was analyzed at 50 ug/ml, followed by biotinylated anti-human IgG (Vector) 5 ug/ml. Binding was detected using Alexa Fluor-647-labeled streptavidin from Molecular Probes (1 ug/ml). As diluent the blocker solution was used.
