Click Preference from File in the main menu.

Users can edit the setting and parameters of Project, Global Setting, Archive, Analyte and Glycan Array Cartoon Options.

Cartoon options for glycan sequences are:

  1. SNFG
  2. CFG
  3. CFG-Hybrid
  4. CFG-BW
  5. Oxford
  6. Oxford-colour
  7. Text

Image style including:

  1. Complete
  2. Normal
  3. Compact

  2. LR: Left to Right
  3. RL: Right to Left
  4. TB: Top to Bottom

Table column selection

Users are able to select the columns to include in the table result view.

  1. Right click on the table header area and select ‘Choose columns’ – Users are able to select columns to include in the table result view.
  2. Items in the ‘Select Columns’ box (right box) are included in the table
  3. Items can be selected by clicking the arrow icons in the middle

Merge multiple data set for comparison

This tool is used for comparing multiple data sets side-by-side.

Click Tools in the main menu and select Merge Glycan Array Data.

  1. Name – A name of data comparison
  2. Add the data sets for the comparison by clicking ‘Add’ button

  1. Alias - Users are able to create a new sample name for data comparison
  2. Fluorophore – Select the fluorophore which is extracted from the original GPR or Proscan file
  3. Scan Power – Select the scan power which is extracted from the original GPR or Proscan file
  4. Statistical Method – Select one from Average or Elimination
  5. Signal Type – Select one from Median-B, Mean-B, Median, Mean, MedianS-B, MeanS-B
    • Median-B: retrieve the intensity value from the ‘Median-B’ (median – background) column in the original GPR or Proscan file
    • Mean-B: retrieve the intensity value from the ‘Mean-B’ (mean – background) column in the original GPR or Proscan file
    • Median: retrieve the intensity value from the ‘Median column in the original GPR or Proscan file
    • Mean: retrieve the intensity value from the ‘Mean’ column in the original GPR or Proscan file
    • MedianS-B: use the intensity value after subtracting the ‘Background’ value from the ‘Median’ value.
    • MeanS-B: use the intensity value after subtracting the ‘Background’ value from the ‘Mean value.
  6. Probe Concentration – Select the probe concentration which is extracted from the Array Data results (tabulations)

Once there are more than two entries, OK button will be activated.

A new table is generated by addressing the intensity values from different experiments on the glycan probes.
Users can export this table into the Excel file for heatmap generation and other data analysis.

Creating heatmaps

Users can generate heatmaps using the exported excel file and heatmap template (Heatmap template_forWin.xlsm or Heatmap template_forMac.xlsm) included in the CarbArrayART software package.

*Please enable the macro of the heatmap template Excel file if you see the security working

  1. Copy the table from the exported excel file and past to the spreadsheet
  2. Click 'Colour Format' button (A)

  3. Click the area selection button (B)
  4. Select items from the table which are used for heatmap calculation.
  5. Click 'Colour format' button (C)

*There are two spreadsheets included (3 coloured and 4 coloured heatmaps)
